Act on muscle contractile mechanisms for muscle spasm.

Centrally acting:
• eg baclofen, carisoprodol, diazepam
• block polysynaptic pathways at spinal cord level

Direct acting:
• eg dantrolene
• interferes with calcium release in muscle fibres
