Zidovudine

Zidovudine (AZT):
• nucleoside reverse transcriptase inhibitor (NRTI)
• analogue if thymidine
• suppresses synthesis if viral DNA after conversion to active form (zidovudine triphosphate)  becomes incorporated into the DNA strand  prevents reverse transcriptase from adding more bases
• indicated for HIV infection; good penetration of CNS when compared to other agents; always used in conjunction with another antiretroviral agent as monotherapy can lead to resistance
• adverse effects – anaemia; neutropenia; lactic acidosis
